S100A4 belongs to the S100 super-family of proteins containing 2 EF-hand calcium-binding domains. S100 genes include at least 25 members, including S100A1-S100A18, trichohyalin, filaggrin, repetin, S100P, and S100Z. S100A4 exerts its function via direct interaction with a number of proteins including P53, P63, non-muscle myosin IIA, 64 integrin, and liprin b1. S100A4 is overexpressed in highly metastatic cancers, which makes it useful as a marker of tumor progression. Recognizes a glycoprotein of 75 kDa, identified as low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). NGFR is expressed in various neural crest cells and their tumors such as melanocytes, melanomas, neuroblastomas, pheochromocytomas and neurofibromas. Reportedly, anti-NGFR is a reliable marker for desmoplastic and neurotropic melanomas. NGFR is expressed in mature non-neural cells such as perivascular cells, dental pulp cells, lymphoidal follicular dendritic cells, basal epithelium of oral mucosa and hair follicles, prostate basal cells, and myoepithelial cells. Anti-NGFR stains the myoepithelial cells of breast ducts and intra-lobular fibroblasts of breast ducts. This antibody reacts with the C-terminus of TGF alpha and shows no cross-reaction with EGF and the neuropeptide synenkephalin. TGF-beta (aa50) is a growth factor with 33% homology to EGF, binds to EGFR, activates tyrosine phosphorylation of the receptor, and stimulates cell proliferation. Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton, and functions in the determination of cell shape, arrangement of transmembrane proteins, and organization of organelles. Spectrins function as membrane organizers and stabilizers, composed of non-homologous alpha and beta chains, which aggregate side-to-side in an anti-parallel fashion to form dimers, tetramers and higher polymers. Spectrin alpha I and spectrin beta I are present in erythrocytes, whereas spectrin alpha II (also designated fodrin alpha) and spectrin beta II (also designated fodrin beta) are present in other somatic cells. Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. Recognizes a protein of 40 kDa, identified as CD7 (Workshop IV; Code T155). CD7 is a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA positive) from pleural mesotheliomas (rarely or weakly CEA positive). This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer. Galectin-13 is suggested to have a developmental role in the placenta and may display immunobiological activity in feto-maternal blood-spaces. This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. This MAb reacts with a protein of 22 kDa, identified as beta sub-unit of FSH. It does not cross react with the alpha sub-unit. Follicle stimulating hormone (FSH) is a hormone synthesized and secreted by gonadotrophs in the anterior pituitary gland. In the ovary, FSH stimulates the growth of immature Graafian follicles to maturation. As the follicle grows, it releases inhibin, which deactivates the FSH production. In men, FSH enhances the production of androgen-binding protein by the Sertoli cells of the testis and is critical for spermatogenesis. FSH and LH act synergistically in reproduction. Recognizes a protein of 205 kDa-220 kDa, identified as CD45RA (Workshop III). CD45RA is isoforms of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4 T-cells, 50% of peripheral CD8 T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. CD45RA and CD45RO define complementary, predominantly non-overlapping populations of resting peripheral T-cells. This MAb is useful in study on the subpopulation of CD4 or CD8 T-cells. MAb VU-11D1 reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. The dominant epitope of MAb VU-11D1 is the 7-mer TSAPDTR of the MUC1 tandem repeat as established with 'epitope fingerprinting'. The'extra cellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion. Gastrin, is a hormone that normally formed by mucosal cells in the gastric antrum and by the D cells of the pancreatic islets. Its primary function is to stimulate secretion of HCl by the gastric mucosa. HCl, in turn, inhibits gastrin formation. It also responsible for stimulating smooth muscle contraction and increasing blood circulation and water secretion in the stomach and intestine. Gastrin is regulated by epidermal growth factor in both mice and humans. Gastrin is excreted in excess by pancreatic tumors in the Zollinger-Ellison syndrome. Recognizes a cell surface protein of 220-240 kDa, identified as L1 cell adhesion molecule. The L1CAM gene, which is located in Xq28, is involved in three distinct conditions: 1) HSAS (hydrocephalus-stenosis of the aqueduct of Sylvius); 2) MASA (mental retardation, aphasia, shuffling gait, and adducted thumbs); and 3) SPG1 (spastic paraplegia). The L1, neural cell adhesion molecule (L1CAM) also plays an important role in axon growth, fasciculation, and neural migration as well as in mediating neuronal differentiation.
